The study identifies a direct mechanism: <strong>increased soluble TNF Receptor-2 (sTNFR2)<\/strong> leads to the <strong>neutralization of inflammatory TNF\u03b1 and TNF\u03b2<\/strong>, reducing the cytokine storm responsible for lung injury.<\/p>\n<p><strong>Clinical Context<\/strong><\/p>\n<p>The potency assay was performed on plasma samples from 24 subjects enrolled in the <em>Phase 1\/2a double-blind randomized controlled trial<\/em> of UC-MSC therapy for COVID-19 ARDS. Samples were collected on <strong>Day 0 (baseline)<\/strong>and <strong>Day 6 (post-treatment).<\/strong><\/p>\n<p><strong>Key Findings<\/strong><\/p>\n<ul>\n<li><strong>sTNFR2 levels increased significantly<\/strong> in the UC-MSC group (26,609 \u00b1 846 pg\/ml vs. 23,111 \u00b1 760 pg\/ml; <em>p = 0.021<\/em>).<\/li>\n<li><strong>TNF\u03b1 decreased<\/strong> from 950 \u00b1 226 pg\/ml to 319 \u00b1 40 pg\/ml (<em>p = 0.048<\/em>).<\/li>\n<li><strong>TNF\u03b2 decreased<\/strong> from 2,944 \u00b1 735 pg\/ml to 810 \u00b1 126 pg\/ml (<em>p = 0.032<\/em>).<\/li>\n<li>Control subjects showed no significant change over the same period<\/li>\n<\/ul>\n<p><strong>Interpretation<\/strong><\/p>\n<p>These results demonstrate that JadiCells <em>induce a regulatory immune phenotype<\/em>, promoting <strong>T-cell quiescence<\/strong> and <strong>regulatory T-cell (Treg) expansion<\/strong> through sTNFR2 signaling.
